Asymbiotic germination in three Chloraea species (Orchidaceae) from Chile Gayana Botánica

ABSTRACT Orchids require symbiotic fungi and/or specific conditions to germinate. Asymbiotic techniques have been shown successful for orchid germination. In Chile, Chloraea include many endemic, and potentially ornamental, terrestrial orchid species. In this study, individuals of Chloraea crispa, C. gavilu and C. virescens were manually autopollinated. The resulting capsules were sterilized and seeds were aseptically obtained. We evaluated asymbiotic germination in: Agar Water (AW), Knudson C (KC), Banana Culture Media (CMB), Tomato Culture Media (CMT), Malmgren Modified (MM), Murashige and Skoog (MS), and MS modified (MS1/2). ABSTRACT The sugar apple is highly dependent on the pollination and fruit tree pollen performance is crucial for successful fertilization and fruit set. The objective of this study was to determine the cardinal temperatures for in vitro pollen grains germination and evaluate the effect of temperature on pollen tube growth of sugar apple. The experimental design was in a completely randomized with eight treatments (10, 15, 20, 25, 30, 35, 40 and 45 ºC), four replicates and each plot was constituted by two slides per parcel. Germinação in vitro e embriogênese somática a partir de embriões imaturos de canela sassafrás (Ocotea odorifera Mez) Rev. Bras. Bot.

Embriões zigóticos em diferentes estádios de desenvolvimento foram utilizados para avaliar o potencial de germinação e competência embriogênica in vitro. Frutos coletados entre 15 e 360 dias após a antese foram avaliados em relação ao diâmetro (mm), comprimento (mm) e massa fresca (g). Para avaliar a percentagem de germinação in vitro, embriões zigóticos em diferentes estádios de desenvolvimento foram inoculados em meio de cultura composto de sais minerais de Murashigue & Skoog (MS), ou em meio MS suplementado com 4,4 miM de BAP, sacarose (3%), ágar (0,6%), carvão ativado (0,15%). The objective of this work was to characterize the morphology and viability of the pollen of 15 genotypes of castor bean (Ricinus communis L.) and to generate information that can assist in the selection of highly promising male parents for future use in genetic improvement programs aimed at producing seeds for oil extraction. Acetolysis and scanning electron microscopy was used to characterize the morphology of the pollen. Morphology and viability of pollen grains from passion fruit species (Passiflora spp.) Acta Botanica

The characterization and viability of pollen grains are useful tools to guide crosses in breeding programs. The objective of this study was to describe the morphological patterns and viability of pollen grains from five accessions of Passiflora edulis f. flavicarpa O. Deg. and five accessions of Passiflora setacea DC. Pollen morphology descriptions were made using light microscopy and scanning electron microscopy, whereas the viability analysis was performed by in vitro germination and histochemical analysis (Lugol's solution and 2,3,5-triphenyltetrazolium chloride). Self-, cross- and interspecific pollinations in Passiflora capsularis and P. rubra Rev. Bras. Bot.

This study aimed to characterize the reproductive system of Passiflora capsularis L. and P. rubra L. In vivo controlled pollinations, in vitro pollen germination and pollen-ovule (P:O) ratio evaluation were conducted. In self-pollination, intraspecific and interspecific pollination, P. capsularis showed means of 62.5, 68.7 and 48.4% of fertilized flowers, while in P. rubra, the averages were 67.2, 62.5 and 46.9%, respectively. For in vitro germination, 52.2% of P. capsularis pollen grains germinated while in P. rubra, the percentage was 64.4. The P:O ratio was 22.4 for P. capsularis, and 27.4 for P. rubra, which included them in the category of obligatory autogamous.
